솔잎추출물의 모발 성장 촉진 효과에 관한 연구

- Abstract
- 본 연구는 솔잎추출물을 이용하여 모발성장 촉진효과에 대해 알아보고자
생체외(in vitro)와 생체내(in vivo)실험을 통해 생리학적 변화와 기능학적
검사인 혈액학적 및 항산화 실험, 조직학적 모발형태 변화, 분자생물학적
발현량을 체계적으로 비교분석하였다. 실험동물은 생후 5주령 된 웅성
C57BL/6 mouse를 이용하였고, 실험군은 E1 도포군(솔잎 2%), E2 도포군
(솔잎 4%) 및 대조군(saline)과 양성대조군(3% MXD)으로 하여 군별 당 5
마리를 이용하여 각 시료에 따라 제모한 후 등 부위에 시료를 1일 2회, 매
회 120㎕씩 주 6일, 4주 동안 경피 도포 후 관찰하여 다음과 같은 결과를
얻었다.
첫째, 솔잎추출물의 전자공여능을 측정한 결과는 BHT와 솔잎추출물 모두
농도가 증가함에 따라 전자공여능이 증가하는 양-반응 관계를 나타났다.
둘째, 적출한 피부 조직의 유해산소 대사 효소 활성 변동의 함량은 대조군
에 비하여 양성대조군과 실험군(E1, E2)이 XO와 TBARS가 각각 통계적으
로 유의하게 낮았고, SOD와 CAT는 대조군에 비하여 양성대조군과 실험군
(E1, E2)이 유의하게 증가하였다. 이를 볼 때 솔잎추출물이 인체에 유해한
활성산소종의 방어기전으로서의 작용됨으로 육모 촉진과 탈모예방에 효과
가 있는 것을 볼 수 있다.
셋째, 생리적 기능에 대한 효능을 확인하기 위한 체중, 식이량, 음수량 및
식이효율의 변화는 경미하게 관찰되었으나 모든 군에서 통계적 유의성은
없었으며, 독성실험의 안전성 지표로 장기의 심장, 비장, 간, 신장(좌·우) 및
흉선 무게를 측정한 결과는 심장, 비장, 간, 신장(좌·우)및 흉선 무게 모두

대조군에 비해 양성대조군과 실험군(E1, E2)에서 통계학적 유의성은 나타
나지 않았다. 본 연구의 결과로 솔잎추출물은 장기기능별에 대한 직 ·
간접적인 영향은 미미한 것으로 나타났다.
넷째, 기초대사성을 보기위해 혈액학적 검사의 백혈구 수치변화에서는 대
조군에 비하여 양성대조군에서는 통계적 유의성이 없었으나, 실험군(E1,
E2)은 대조군에 비해 통계학적 유의성 있게 나타났다. 실험군 E1에서는 백
혈구 중 염증관련 세포인 호중구에서 유의하게 증가하였다. 단핵구, 호산구,
호염기구, 적혈구, 혈색소, 적혈구 용적, 혈소판 수치는 모든 실험군 간에
서 통계적 유의성이 나타나지 않았다.
다섯째, 피부 조직을 H&E로 염색하여 광학현미경으로 관찰한 결과 100배
율에서 육안으로 확인된 모낭의 수, 모낭깊이, 진피의 두께 모두 대조군에
비해 양성대조군, 실험군(E1, E2)에서 통계적으로 유의하게 증가하는 경향
이 확연하게 보였다. 이를 볼 때 솔잎추출물이 모발 성장 촉진효과가 있는
것을 볼 수 있었다.
여섯째, 모발성장을 유도하는 중요한 지표로 간주되는 피부조직 내 효소
활성도를 측정하기 위해 ALP의 효소 활성도 측정 결과는 대조군에 비해
모든 군에서 증가하는 경향을 나타내어 통계적 유의성을 보였다. 증식과
분열이 활발한 세포에서 많이 발현되어지는 γ-GT가 대조군에 비해 모
든 군에서 증가하는 경향을 나타내어 통계적 유의성을 보였다.
일곱째, 피부조직 내 IGF-1 mRNA의 성장인자 발현량은 모발성장과 함께
증가하는 경향을 보였으며, 대조군에 비해 모든 군에서 통계적으로 유의성
있게 증가하여 높은 활성을 보였고, 피부조직 내 VEGF mRNA의 발현량
또한 실험군 E2> 실험군 E1> 양성대조군> 대조군 순으로 높은 활성을
보 였고, 저해인자인 TGF-β1 mRNA의 발현량은 대조군에 비하여 양성
대조군, 실험군(E1, E2)에서 많이 줄어들어 유의성이 나타났다
