Association between CR4(CD11c/CD18) gene Polymorphisms and Behcet's disease
- Abstract
- Objectives: Behcet's disease (BD) is a chronic and multisystemic inflammatory disease. The pathogenesis cause of BD is still uncertain, but the interaction between leukocyte adhesion and integrins, leukocyte migration into inflammatory site are important responses in the development of inflammatory in BD. Complement receptor type-4 (CR4, CD11c/CD18) is a member of the β2-integrins and plays important role in leukocyte migration into the inflammatory site. This study investigated CR4 (CD11c/CD18) polymorphisms association with BD.
Methods: The single nucleotide polymorphisms (SNPs) in the CD11c (rs2230429C>G, rs2929G>A) and CD18 (rs2070946A>G, rs235326T>C, rs760456C>G, rs684G>A) were genotyped using PCR-RFLP among 305 BD patients and 266 healthy controls in Korean.
Results: The homozygous CD11c rs2230429C/C genotype and rs2929G/G genotype were significantly higher in BD patients than in healthy controls (p = 0.0004, OR = 1.8, 95% CI = 1.31-2.56; p = 0.021 OR = 1.8, 95% CI = 1.06-2.05, respectively). The homozygous CD18 rs235326 C/C genotype was significantly higher in BD patients than in healthy controls (p = 0.002, OR = 1.7, 95% CI = 1.20-2.36). The allele frequencies of CD11c rs2230429 C (p = <.0001, OR = 1.7, 95% CI = 1.31-2.26), rs2929 G (p = 0.013, OR = 1.4, 95% CI = 1.07-1.78) and CD18 rs235326 C (p = 0.001, OR = 1.6, 95% CI = 1.20-2.10) were associated with the increased risk of BD. There was no significant difference in the genotype and allele frequency of the CD18 rs2070946A>G, rs760456C>G and rs684G>A.
Conclusions: These results suggest that the CD11c Pro517Arg (rs2230429), 3’UTR (rs2929) and CD18 Val441Val (rs235326) polymorphisms are associated with BD.
